High counting rates from 125I in radioimmunoassays.

چکیده

Responses from one manual and four automatic gamma counters for assaying (125)I were compared. The resolution times quoted by manufacturers when applied in a standardized correction formula to obtain true counts from observed counting rates above 10(4) counts per second appeared to be inadequate. The need to investigate the errors associated with high counting rates for all instruments and electronic settings employed to determine (125)I in radioimmunoassays is emphasized.

3H and 125I radioimmunoassays of haloperidol compared with fluoroimmunoassay involving antibody coupled to magnetizable solid phase.

Radioimmunoassays for haloperidol are described, involving use of tritium- or 125I-labeled drug or tritium-labeled spiroperidol, and a rabbit antiserum to a drug/bovine serum albumin conjugate. The 125I-labeled drug was prepared by the Chloramine T iodination technique. A fluoroimmunoassay for haloperidol is also described in which the antiserum is coupled to magnetizable solid-phase medium, an...

Radioiodinated lipoproteins: absorption of 125I radioactivity by high density solutions.

Concentrations of potassium bromide commonly used for separation of lipoproteins were shown to cause absorption of 125I and thus reduce the counting efficiency of the labeled lipoproteins. Chloroform was shown to cause a 50% reduction in counting efficiency of lipid from 125I-labeled lipoprotein. No reduction of counting efficiency was observed in the presence of high density solutions when 131...

Internal sample attenuator counting (ISAC). A new technique for separating and measuring bound and free activity in radioimmunoassays.

I describe a new method for the separation and counting of bound and free activity in radioimmunoassays. Particles containing a radiation-absorbing (attenuating) material are added to the assay. They shield the radiation from either the antibody-bound or the free radioligand. This obviates such manipulations conventionally involved in the separation and counting steps of radioimmunoassays as ce...

Anti-triiodothyronine autoantibodies interfere with two recently commercialized radioimmunoassays for free triiodothyronine.

We examined the effect of anti-triiodothyronine autoantibodies on results of RIA of free triiodothyronine with two commercial kits (Amersham International and Diagnostic Products Corp.). Both methods gave falsely high results because the 125I-labeled analogs used in these RIAs were bound by the autoantibodies.

Counting Non-Aqueous Samples in the TopCount® Microplate Scintillation and Luminescence

The microplate format has become a standard for many biological assays such as high throughput screening for drug discovery and radioimmunoassays. Productivity with these high volume procedures has been limited by the amount of sample handling required and low sample counting throughput. Now, however, microplate assays can be efficiently counted in microplates with the Packard TopCount Micropla...
